The topic of our round-table workshop May 29, 2025 was i-HMBC Methodology: Applications and Enhancements
In 2023, researchers at Merck & Co. Inc. reported the i-HMBC methodology which allows differentiation of 2JCH from 3+nJCH correlations based on comparative analysis of two-bond 2Δ1H(13/12C) and three-bond 3Δ1H(13/12C) isotope shifts. This innovation obviates the sensitivity challenges associated with the 1,1-ADEQUATE experiments and supplants the structural characteristics required for confident analysis by the novel SEA-XLOC pulse sequence. While proven to be effective in the solution of complex structure problems, hurdles associated with data acquisition and processing have hindered widespread adoption of the i-HMBC strategy. In this presentation, we will discuss the foundations of the method, pulse sequence selection, and optimized approaches to data processing and analysis. These new improvements will be demonstrated on model compounds and real-world applications alike.

Thank you for this webinar. Being able to distinguish 2-bond from 3-bond correlations is so essential for the confirmation of structure. Thanks for clearly explaining this new technique.
